Influence of PAS on the Proteolytic Action of Pepsin

Abstract
|
|
|
Experiments were carried out to observe the influence of PAS on the proteolytic action of pepsin.
To determine the effect of PAS,
1ml of pepsin solution(15mg/ml) was placed in a test tube at various pH and either 1ml, of water or PAS solution (15mg/ml) was added and the contents mixed by swirling after which 1ml of Human Serum Albumin was added.
Following an incubation period of 30minutes 10ml of 10 %Trichloracetic Acid was added to stop the reaction.
After removing supernatant, 2cc of 0 2N NaoH was added in test tube and 8cc of Biuret Reagent was added. Heparin solution (10mg/ml) is known as the inhibitant of pepsin activity and same test was done with heparin solution to compare the influence of PAS and heparin on the proteolytic action of pepsin.
The electrophotometric reading showed heparin inhibited the proteoly tic action at optimum pH of pepsin activity but PAS did not.
